流式细胞基本结构
流式检测细胞仪由三部分企业构成
1.流量系统,包括流室和流量驱动系统;
2.光学信息系统,包括可以激发光源和光束收集管理系统;
3.电子系统,包括光电转换器和数据处理系统。流式细胞仪通过将荧光标记的细胞或悬浮在液体中的颗粒一个接一个地通过一个样品池,同时将荧光信号可以通过进行荧光探测器捕获并转换成一个代表前向散射角、侧向散射角和不同导致荧光材料强度的电脉冲信号,分析直方图和三维结构图。
用于FCM的样品是单细胞悬浮液,例如血液、悬浮细胞培养基、各种体液、新鲜实体肿瘤的单细胞悬浮液和石蜡包埋组织的单细胞悬浮液。
流式细胞基本原则
1.使各种离子液体和悬浮培养细胞研究样本企业新鲜,尽快实现完成一个样本可以制备和检测;2.针对问题学生通过不同的细胞可以作为研究样本数据信息进行分析选择适当提高洗涤、酶消化或EDTA处理,以清除杂质,使粘附的细胞之间彼此相互分离而形成具有一种单细胞状态;3.对于新鲜信息的实体肿瘤组织,可以选择或结合酶消化、机械破碎和化学结构分散来获得足够重视的单细胞悬液;4.对石蜡包埋可以组织应先切成若干40-50um厚的蜡片,经二甲苯脱蜡到水后,再用前述研究分析教学方法发展以及技术制备得到单细胞悬液;5.单个细胞悬浮液中的细胞数目不应小于10.000.
流式细胞技术的应用:
1.细胞内DNA测量的变异相关系数进行较小,一般都是小于2%以下;
2.可以准确地进行DNA倍体分析;
3.利用荧光染料对细胞内蛋白质和核酸进行定量分析;
4.快速进行细胞分选和细胞收集;
5.医学技术应用:通过各种干细胞测定研究免疫系统功能,癌患者的多重耐药性研究,细胞功能和代谢动力学,可分析的血小板(心血管疾病),流式细胞术和分子结构生物学的教学和研究;
6它可应用于外周血内皮细胞的测定和调节性T细胞。
