免离心技术是利用物体高速旋转时产生的强大离心力,使置于旋转体中的悬浮颗粒沉降或漂浮,从而使部分颗粒浓缩或与其他颗粒分离。这里的悬浮粒子通常指悬浮的细胞、细胞器、病毒和生物大分子。当转子高速旋转时,当悬浮颗粒的密度高于周围介质的密度时,颗粒远离轴而沉降。如果粒子的密度低于周围介质的密度,则粒子向轴移动并漂浮。常用的离心机有很多种。一般低速离心机高转速小于6000rpm,高速离心机高转速小于25000rpm,超速离心机高转速大于30000rpm。
免离心法是通过离心机的高速运转,使免离心加速度超过重力加速度几百倍或几千倍,从而提高沉降速度来加速药液中杂质的沉淀和去除的方法。其原理是利用混合液体的密度差来分离料液,比较适合分离含有细小颗粒或絮状物的难以沉降和过滤的悬浮液。离心机是利用电机的高速旋转产生离心力场,根据不同物质之间密度、形状、大小的差异,分离提取混在悬浮液中或乳化液中不相溶的不同颗粒的仪器设备。它应用于生物、医学、化工等领域。利用离心机分离、纯化和提取样品的技术称为离心分离技术。常用的离心技术有:沉降离心法、差速离心法、密度梯度离心法、分析超速离心法、离心淘洗法、区带免离心法和连续流动离心法,其中实验室常用的有沉降离心法、差速免离心法和密度梯度离心法。其他技术需要特殊的离心机或转子。
