随着测序技术的飞速发展,引起生物领域的巨大变革。以前,需要从大量细胞获取足够多的DNA 进行测序,因此测序结果是这些 “整体”的表征。然而,由于异质性,相同表型的遗传信息可能存在显 著性差异,很多低丰度的内容会在整体表征丢失。为了弥补传统高通量测序的局限性,单细胞测序应运而生。
生命的发展是自然界的非凡壮举之一。尽管经过几十年的钻研,生物学家仍然无法完全理解这一过程。首先,测序不仅仅是对一个测序,而是可以对单个的基因组或转录组测序。
单细胞测序自2009年问世,越来越多地被应用在科研领域。2015年以来,10X Genomics、Drop-seq、Micro-well、Split-seq等技术的出现,降低了成本门槛。自此,被广 泛应用于基础科研和临床研究。许多领域都占有一席之地,对于癌病早期的诊断、追踪以及个体化治疗具有重要意义。
传统方法显示的信息也是多水平的平均,而单细胞测序可以完整反映同一群体中不同的基因组和转录组状态。出现解决了从混合样本筛选异质的问题,成熟应用必将生命科学向前迈进一大步。那么是如何实现的呢?以单细胞RNA-seq为例,我们简单介绍一下原理:
1.分离,分别构建测序文库,测序。这种想法的通量很低,成本很高。
2.基于条形码识别。其核心思想是在对其mRNA测序前进行逆转录时,给每个添加一个独特的标签序列。即使是混合测序,也可以把条形码相同的片段视为来自同一个。通过这种策略,我们可以通过一次建立一个数据库来测量上万个单一的信息。
3.基本数据处理过程是高维矩阵(有很多行和列)。因此,对处理算法和工具的要求比一般的批处理排序更高。另外,通过数据的特性,还可以进一步挖掘现有大量测序中无法获得的信息。因此,单细胞测序技术的发展是必要的。
